Vaccinations are typically intramuscular not intravenous (an mRNA vaccine can do nothing if not deposited in cytoplasm).

Alveoli or myocyte, the spike protein gets presented to the adaptive immune system. The difference is that with an mRNA vaccine in a myocyte, there is no viral replication.
